Cy3-UTP(Cyanine 3UTP) can replace UTP as a substrate for T7 RNA polymerase and generate labeled probes through in vitro transcription. The resulting assembled mRNA will emit orange fluorescence with the involvement of Cy3-UTP. In addition, Cy3labeled mRNA is readily visible under ultraviolet light without further staining after gel electrophoresis.Probes generated by these methods are suitable for multicolor fluorescence analysis, specifically dual-color expression arrays in conjunction with cyanine3-UTP .
